Dual Integrated Light Scattering system dedicated to protein characterization. Use as a chromatography detector with any GPC/SEC system to give absolute size and absolute molecular weight. Use in cuvette mode for the detection of large aggregates.

Overview

Dynamic light scattering technology (DLS) used to measure molecule size, and Static Light Scattering (SLS) to determine molecular weight.

  • Use as a chromatography detector to measure absolute size and absolute molecular weight, aggregation and conjugation by connecting the Zetasizer µV to your GPC/SEC system and concentration detector, using standard chromatography tubing.
  • Use in cuvette mode - simply remove the flow cell from the optics unit and insert a cuvette containing the sample.
  • Switch in seconds from chromatography detector to cuvette mode.
  • Accurate and repeatable results using precise temperature control.
  • Sensitivity – 0.1mg/mL 15kDa protein (Lysozyme).
  • Only 2µL sample required in cuvette mode to preserve sample.
  • 8µL flow cell for chromatography mode ensures minimal band broadening to preserve the resolution of the separation.
  • Connect to any Viscotek system for full software integration and truly automated operation, or third party SEC system using OmniSEC software and OmniFACE interface.
  • Automated temperature trends for aggregation temperature determination (melting point).

How it works

The Zetasizer µV incorporates two techniques that can be used in both chromatography detector mode and cuvette mode.

Dynamic Light Scattering is used to measure particle and molecule size. This technique measures the diffusion of protein molecules moving under Brownian motion, and converts this to size and a size distribution using the Stokes-Einstein relationship.

Static Light Scattering is used to determine molecular weight. When used as a detector, the light scattering intensity in combination with the signal from a concentration detector enables the absolute molecular weight to be calculated.

In cuvette mode, the intensity of a few concentrations are measured to enable a Debye plot to be constructed. From this a molecular weight and second virial coefficient can be calculated.

The software is designed to be versatile and rich in features without compromising ease of use. Standard Operating Procedures (SOP) simplify routine measurements, a quality factor gives a single place for confirmation of data quality, and an expert advice system is provided to assist with data interpretation.
