Combining SEC with the Zetasizer Nano as a dynamic light scattering detector
Size Exclusion Chromatography (SEC) is a common technique in biochemistry laboratories and routinely used for purification, identification, and quantification of protein mixtures. Dynamic light scattering (DLS) is traditionally used for pre-column size & polydispersity measurements, along with aggregate quantification.
Aside from the enhanced sensitivity, a big advantage of dynamic light scattering coupled with SEC is the ability to perform Absolute SEC (ASEC), or size exclusion chromatography in the absence of costly and time consuming column calibration. Dynamic light scattering provides a direct measure of the protein size, as opposed to the SEC derived size extracted from a calibration curve. In addition, size vs. mass relationships for globular proteins and polymeric systems are well known, and can be used to calculate the molecular mass from the dynamic light scattering measured hydrodynamic radius.

Determination of protein structure is a priority for understanding the role and function of many proteins. One indicator of structure is the size of the molecule. This rapid method is a convenient route to obtain an indication of the quaternary structure of a protein. Many proteins show profound changes in different solution conditions, with the most extreme structure change being protein denaturation. Similar structure changes may happen as a function of time, pH, ionic strength and changes in various other solution conditions. Dynamic light scattering is an ideal protein characterization tool to quickly assess the condition of the molecule in solution under such a range of conditions.
